چکیده

1369 Eszopiclone and Sleep Spindles in Schizophrenia—Wamsley et al INTRODUCTION Cognitive deficits are the strongest predictor of functional outcome in schizophrenia,1 and antipsychotic drugs (APDs) are relatively ineffective in treating them (e.g., Sergi2). Amelioration of cognitive deficits has thus become a priority of the schizophrenia research community and is the focus of largescale studies.3 A limitation of these efforts is that cognition is measured in cross-section. While this provides a valid snapshot of function, it misses critical aspects of learning and memory that happen offline, over time and with sleep. Following encoding, memories undergo “consolidation” processes that stabilize, enhance, integrate, and reorganize memory in the brain. A wealth of evidence from molecular, cellular, neural network, regional brain activation, and behavioral studies of birds,4 rodents,5 cats,6 and humans7 demonstrates that sleep plays a critical role in memory consolidation. This work suggests an evolutionarily conserved function for sleep in memory consolidation. In schizophrenia, there is evidence of both abnormal EFFECTS OF ESZOPICLONE ON SLEEP AND MEMORY IN SCHIZOPHRENIA

Effects of eszopiclone and zolpidem on sleep and waking states in the adult guinea pig.

STUDY OBJECTIVE The present study was designed to compare and contrast the effects of eszopiclone and zolpidem on the states of sleep and wakefulness in chronically instrumented, unanesthetized adult guinea pigs. DESIGN Adult guinea pigs were implanted with electrodes to record sleep and waking states and to perform a frequency analysis of the EEG. Eszopiclone (1 and 3 mg/kg) and zolpidem (1 ...

Melatonin for Olanzapine Metabolic Effects

A study found that patients taking the recommended 3-mg dose of the sleep aid eszopiclone (Lunesta) at bedtime continued to have impaired driving skills, memory, and coordination for as long as 11 hours after ingestion. Patients were often unaware they remained impaired. As a result, the FDA has recommended decreasing the starting dosage of eszopiclone to 1 mg at bedtime. The change should resu...

Eszopiclone: its use in the treatment of insomnia

Eszopiclone is the S-isomer of racemic zopiclone, a cyclopyrrolone with sedative-hypnotic activity that has been available in Europe, Canada, and Latin America since 1987. Eszopiclone acts by binding to the GABA(A) receptor. In contrast to the benzodiazepine (BZD) hypnotics, eszopiclone has more selectivity for certain subunits of the GABA(A) receptor. Oral eszopiclone is rapidly absorbed and e...

Management of insomnia in elderly patients using eszopiclone

Insomnia is a common sleep complaint in the elderly. The safety and efficacy of eszopiclone, a non-benzodiazepine hypnotic, in elderly patients with chronic insomnia has been established in two 2-week and one 12-week randomized, double-blind, placebo-controlled trials. Eszopiclone 1 mg was effective in reducing sleep latency. Eszopiclone 2 mg was effective in reducing latency to sleep and for i...

Residual effects of eszopiclone and placebo in healthy elderly subjects: a randomized double-blind study

Next-day residual effects are a common problem with current hypnotics. The purpose of the present study was to evaluate the residual effects of eszopiclone on the physical and cognitive functions of healthy elderly people in the early morning and the day following drug administration. Four men and six women aged 63-72 years were administered eszopiclone 1 mg or placebo in a randomized, double-b...
